Wolfgang Liedtke

Wolfgang Liedtke is a neurologist, translational neuroscientist and Professor of Neurology at Duke University with over 15 years of clinical development experience spanning discovery research through late-stage drug development. He has led global neurology programmes at Regeneron and Anavex, including registrational Alzheimer’s disease trials, with extensive FDA and EMA engagement. His expertise spans biomarker-driven clinical development, translational neuroscience, neurogenetics, precision medicine and neuroimaging approaches to accelerate neurodegenerative therapeutics development. He discovered the TRPV4 ion channel while a scholar at Rockefeller University, and foundational mechanisms of neuronal chloride regulation in his lab at Duke, where he provided clinical care for more than 5000 patients.

- Moving beyond single-modality endpoints by integrating structural and functional neuroimaging (MRI, PET) with remote digital biomarkers (EEG, actigraphy, speech and cognitive sensors) to better capture neurodegenerative disease biology and treatment response
- Building a regulator-facing multimodal endpoint framework through practical harmonisation of imaging and digital biomarker platforms, with cross-validation against fluid biomarkers such as p tau217 and NfL to strengthen biological plausibility and regulatory confidence
- Applying lessons from Phase IIb/III Alzheimer’s programmes, EMA CHMP/ SAG-Neurology engagement, and ALS and rare disease trials to improve patient stratification, reduce screen failures, and create actionable endpoint strategies across AD, ALS, PD, and rare CNS indications
